霍亂毒素

由霍亂弧菌產(chǎn)生的霍亂毒素（CT）是一種多聚體腸毒素，可將ADP-核糖轉(zhuǎn)移至G蛋白，將腺苷酸環(huán)化酶鎖定在“開啟”位置。

霍亂毒素如何引起腹瀉？通過與腸細(xì)胞膜結(jié)合，霍亂毒素刺激細(xì)胞腺苷酸環(huán)化酶系統(tǒng)，引起氯離子和碳酸氫根離子的過度分泌，這導(dǎo)致增加的液體分泌和霍亂疾病的嚴(yán)重腹瀉特征。CT的五聚體B亞基與GM1單唾液酸神經(jīng)節(jié)苷脂細(xì)胞膜受體結(jié)合，存在于許多細(xì)胞類型中，允許其在細(xì)胞培養(yǎng)中實(shí)驗(yàn)性使用。結(jié)合后，毒素被內(nèi)吞并逆行進(jìn)入內(nèi)質(zhì)網(wǎng)，其中酶活性A亞基（CTA1）易位至胞質(zhì)溶膠。在胞質(zhì)溶膠中，CTA1催化Gs蛋白的ADP-核糖基化，其激活腺苷酸環(huán)化酶，并因此增加cAMP的細(xì)胞內(nèi)濃度。
霍亂毒素如何用于疫苗佐劑開發(fā)？ 與疫苗佐劑的開發(fā)相關(guān)，CT有效地調(diào)節(jié)哺乳動(dòng)物免疫系統(tǒng)并充當(dāng)共同施用的抗原的佐劑。
